Abstract

(±)5,7,4′-Trihydroxyflavanone-5-glucoside-[2- 14 C] was synthesized and the diastereomeric (+) and (-) glucosides were separated by paper chromatography. In buckwheat seedlings the incorporation rate of the (-) enantiomer into quercetin was 16 times higher and into cyanidin 3.6 times higher than that of the (+) compound. In chana seedlings (Cicer arietinum L.) the (-) compound was incorporated into biochanin A (5,7-dihydroxy-4′-methoxy-isoflavone) 14 times better than the (+) compound, whereas in agreement with earlier results the incorporation of both enantiomers into formononetin (7-hydroxy-4′-methoxy-isoflavone) was very low. The results prove the stereospecific incorporation of the naturally occuring (-) · 2 · S · 5.7.4′-trihydroxyflavanone into flavonoids and isoflavonoids. The incorporation of the (+) enantiomer is probably due to a racemization of this compound via the chalcone.

Abstract

Nach Verfütterung von Shikimisäure-U- 14 C an Sprosse von Catharanthus rosens konnten radioaktives Catharantin und Vindolin isoliert werden. Vindolin wurde zur Pikrinsäure = I sowie zum ind-N-Methylnorharmin = II abgebaut. I besaß 95.5% und II 88% der spezifischen Aktivität des eingesetzten Vindolins. Somit ist gezeigt, daß Shikimisäure in den Indolteil des Vindolins inkorporiert worden ist aber nicht in das C 10 -Fragment.

Abstract

Derivatives of adenosine and uridine, which were substituted only at the sugar moiety and which were soluble in CCl 4 , have been prepared. The concentration-dependencies of the IR-spectra of 2′.3′-Isopropylidene-5′-trityl-adenosine (ITA) and 2′,3′-Isopropylidene-5′-trityl-uridine (ITU) in CCl 4 indicated that hydrogen-bonded, cyclic dimers were formed at low concentrations. In solutions of equimolar amounts of ITA and ITU the hetero-associate containing an adenine-uracil-pair was found to be more stable than the homo-associates.

Abstract

Free ethanolamine has been found in leaves of 41 plants. It was detected by means of high voltage electrophoresis and chromatography on thin layers of cellulose. Within 6 hours no conversion of serine to ethanolamine could be observed.

Abstract

Das reversible Bestrahlungsprodukt der Cytidylsäure konnte durch Dünnschichtchromatographie an Polyäthylenimin-Cellulose aus der Bestrahlungslösung abgetrennt und durch Vergleich mit dem auf chemischem Wege dargestellten Produkt (Umsetzung der Cp in wäßriger Lösung mit Br 2 und anschließende katalytische hydrierende Debromierung) als 4-Hydroxy-dihydrocytidylsäure identifiziert werden. In analoger Weise gelang auch die Synthese des 4-Hydroxy-dihydrocytidins. Die Kinetik des Zerfalls der reversiblen Bestrahlungsprodukte von Cn, C und Cp wurde untersucht. Dabei zeigte sich, daß die Beständigkeit wie bei den Uracil-Derivaten in der Reihenfolge Cn, C, Cp - also in Richtung Nucleinsäure - wächst. Durch Verwendung einer besonderen Bestrahlungsanordnung (Hg-Niederdruckbrenner NK 25/7 der Quarzlampen-Gesellschaft, Hanau, mit einer 0,1-m. wäßrigen KBr-Lösung als Filter) gelang es, auch das allerdings sehr instabile reversible Bestrahlungsprodukt des Thymins nachzuweisen.

Abstract

Ribonuclease A was irradiated in water with 60 Co gamma radiation, and the products formed were separated according to their molecular weight by column chromatography on Sephadex G-50. When the irradiations are carried out in nitrogen atmosphere aggregation to dimers and higher polymers is observed. An appreciable fraction of this aggregated component retains enzymatic activity. At higher doses the enzymatic activity of the aggregates is inactivated at the same rate as monomer ribonuclease A. Irradiation in air yields two components; one is equivalent to that found in nitrogen atmosphere, the other has an apparent molecular weight of about 20 000 but contains no enzymatic activity. This last component is not observed when methionine is present during irradiation. In methionine containing solutions all inactivited ribonuclease molecules exist in the form of dimers and polymers. Irradiation in the dry state leads to the same result. Consequently, every model designed to describe the radiation action on ribonuclease has to consider the fact that in solution, in the presence of a protecting agent, and in the dry state the loss of enzymatic activity is always accompanied by aggregation to products with increased molecular weight.

Abstract

Normal and precancerous livers as well as primary and transplanted hepatomas are studied for glycogen content, hexokinase- and glucokinase activity, at the same time measuring glycolysis in the high-speed supernatants of these tissues with different substrates and additives. Experimental results show that: 1) normal and precancerous rat livers have low hexokinase activity and low glycolysis, but high glucokinase activity and high glycogen content; 2) primary DAB hepatomas. on the contrary, have high hexokinase activity and raised glycolysis as well as low glycokinase activity and low glycogen content; 3) transplantable DAB- as well as DENA-hepatomas have high hexokinase activity and high glycolyses. while neither glucokinase activity nor glycogen content have been found; 4) when using glucose as substrate and adding exogenous hexokinase. there is a considerable increase in formation of lactic acid in all tissue supernatants investigated; 5) if glucose-6-phosphate is used instead of glucose, a considerable increase of glycolysis is noted as well. It is concluded from the results obtained that the hexokinase reaction limits the extent of glycolysis in these tissues.

Abstract

Herbizide vom Benzimidazol- und Imidazol-Typ, insbesondere 2-trifluormethyl-substituierte Benzimidazole. sind Hemmstoffe des photosynthetischen Elektronentransportes im Bereich der 2. Lichtreaktion. Die photosynthetische O 2 -Entwicklung in einer Hill - Reaktion mit Kaliumferricyanid als Acceptor wird von einigen Substanzen noch bei einer Konzentration von <10 -6 -m. gehemmt. Bei höheren Konzentrationen wird auch die cyclische Photophosphorylierung unterbunden (Entkopplung der ATP-Bildung). Es ergibt sich eine Abhängigkeit zwischen der Struktur der Benzimidazole und ihrer Hemmwirkung in photosynthetischen Reaktionen: zwischen dem pl 50 -Wert der Hemmung der O 2 -Entwicklung und einer Funktion, die den Verteilungskoeffizienten (Oktanol/Wasser) und den P KA -Wert der Benimidazole enthält, besteht eine lineare Beziehung. Diese Funktion beschreibt auch eine lineare Struktur-Aktivitäts-Beziehung für die Entkopplung der Photophosphorylierung durch Benzimidazole. 2-n-Undecyl-benzimidazol ist ein wirkungsvoller Entkoppler (7,3 · 10 -6 ) der cyclischen und der offenkettigen Photophosphorylierung.

Abstract

After erythrocytes had been treated with NDV, antigens of the Forssman and mononucleosis type could be demonstrated on their surface. The Forssman antigen was found to be either transfered by the virus to the surface of the erythrocytes or unmasked by splitting off sialic acid from chicken red cells. The mononucleosis antigen could only be transfered to the erythrocytes by NDV. Both antigens demonstrable in NDV, proved to be parts of the normal, host-specific component of the virus. The mononucleosis antigen of NDV was similar to that in bovine erythrocytes, but different from the one in sheep red blood cells.

Abstract

Suspended tissue culture cells incorporate 32 P within 60 -90 min. into two high molecular weight RNA fractions which can be separated by chromatography on methylated serum albumin celite columns. One fraction contains mainly ribosomal precursor RNA. the other ribosomal precursor RNA and cell messenger RNA. Under unfavorable growth conditions which do not allow cells to multiply the synthesis of ribosomal precursor RNA is inhibited and high molecular weight RNA accumulates which resembles cellular DNA in nucleotide composition. The experimental results indicate that the synthesis of ribosomal precursor RNA and of messenger RNA in mammalian cells are independently regulated.

Abstract

Bei der Suche nach einem Anreicherungsverfahren für biochemische Mangelmutanten von Hefen wurden mehrere Agenzien ausgetestet. Nur mit Penicillin konnten Anreicherungseffekte erzielt werden. Der Anteil der Mutanten stieg durch die Penicillinbehandlung von 10% nach der Methansulfonsäureäthylester-Induktion auf 20 - 50 Prozent. Veränderungen in der Häufigkeit der Mutantentypen mindern den Nutzeffekt des Anreicherungsverfahrens.

Abstract

The in vivo and in vitro uptake of 22 Na from the hemolymph into cell nuclei of larval salivary glands was measured and compared. The uptake of 22 Na in vivo follows approximately a saturation curve. The respective in vitro curve has a much steeper slope during the first 8 min and this phase is followed by an interval during which the 22 Na content of the nuclei decreases (8 to 16 min). After the 16th min it increases again to reach a nuclear 22 Na content approximately twice as high as that of nuclei in vivo at 60 minutes. The uptake curves are discussed in relation to recent findings on the induction of puffs in polytene chromosomes by inorganic ions.

Chloroplasts oxidize Mn 2⊕ in the light under aerobic conditions. The reaction is independent of the produced hydrogenperoxide. The oxidation is not mediated by the enzyme peroxidase. but by a direct reaction of Mn 2⊕ with an electron carrier which is located between water and the pigment complex II.
