Abstract

The proton chemical shifts of aromatic compounds are strongly concentration dependent. Moreever, for molecules with a dipole moment a temperature dependence of the proton chemical shifts is observed. For hemellitone, p-methylanisole, o-chlortoluene, p-chlortoluene, pyrrole and N-methyl-pyrrole the enthalpies of a dipole-dipole association between -0,7 and -1,8 Kcal could be estimated by NMR measurements combined with the temperature dependence in the whole range of the molefraction (0 → 1).

Abstract

A electric field induced linear and quadratic change in the optical absorption of a chlorophyll a and chlorophyll b monolayer is reported. The chlorophylls are incorporated in a fatty acid monolayer condenser which allows the application of external field strength up to 4 · 10 6 V/cm. The spectrum of the quadratic electrochromic effect shows evidence of different species of aggregates of chlorophyll a and of different behaviour of aggregation of chlorophyll a and b.

Abstract

The structure of radicals in X-irradiated polycrystalline phosphonic acid esters (trichlorphontype) and trichlorphon single crystals has been investigated by ESR. At room temperature in trichlorphon the radical (R0) 2 > P(O) - CHOH - ĊCl 2 (R = CH 3 ) is formed by removal of a chlorine atom. The g-factor is 2,010 and the hyperfine parameters are α H β = 7 ± 0,5 G for the isotropic β-proton and α CL = 7 - 14 G for the two anisotropic chlorine atoms. The structure of this radical type is not influenced by a substitution of the methyl groups by larger alkyl groups (R = C 2 H 5 , C 3 H 7 , C 4 H 9 etc.). ESR-spectra of some other irradiated insecticides are described. Difficulties in the analysis of ESR spectra of aliphatic radicals with chlorine-hyperfine structure (especially quadrupole-effects) are discussed.

Abstract

The formation and conversion of primary radicals in the irradiated phosphonic acid ester trichlorphon and some related compounds at 77 ° K have been studied by ESR. The irreversible changes of spectra by annealing of the sample demonstrate that the chlorine-containing radical type R-CCl 2 1 formed at room temperature represents a secondary type having a precursor with a short half life in the form of a primary radical of similar structure but different conformation which is stable from 77°K to 245°K. A further radical formed at 77°K in trichlorphon is This type disappears at -60°C. In related compounds (DDVP and the acetic acid ester of trichlorphon) a similar radical CH 2 OR is induced, which in contrast to trichlorphon is converted into an oxygen radical with g=2,0076 and g= 2,017.

Abstract

Der native Zustand der Myoglobinmoleküle in wäßriger Lösung ohne Salzzusatz bleibt zwischen pH 7 und pH 4,6 unverändert. Durch HCl-Zugabe erfolgt bei pH 4,6 bis 4,1 eine schwache Assoziation ohne eigentliche Konformationsänderung. Das mittlere Mol.-Gew. steigt bis auf das 1,5-fache des ursprünglichen Wertes. Diese Assoziation erreicht bei pH 4,1 ein Maximum und geht unterhalb dieses pH-Wertes wieder zurück. Bei etwa pH 3,7 beginnt dann die Auffaltung der nativen Struktur zum Knäuel und die Ablösung des Hämins. Unterhalb von pH 2 wird eine mit abnehmendem pH zunehmende Assoziation beobachtet. Wenn die Säuredenaturierung bei der Ionenstärke 0,1 durchgeführt wird, erfolgt die erste Assoziation (pH 4,6 bis 4,1) in verstärktem Maße: Das mittlere Mol.-Gew. steigt auf das 12-fache des nativen Wertes. Der Rückgang der Assoziation bei weiterem Ansäuern ist jetzt unvollständig. Der Salzzusatz bewirkt außerdem, daß die Auffaltung der Molekülsltruktur und die Ablösung der Hämgruppe schon bei pH 4,3 stattfinden. Die Konformationsänderungen des Myoglobins bei der Säuredenaturierung sind reversibel. Die Rekombination zwischen Apomyoglobin und Häminchlorid erfolgt sehr langsam, wahrscheinlich, weil in dem in Frage kommenden pH-Bereich Häminchlorid nicht molekulardispers, sondern kolloid gelöst ist.

Abstract

The diphosphines CCl 3 (Br) P 2 (Br) CCl 3 and CCl 3 (Br) P 2 Br 2 are formed when solutions of P 4 in CCl 3 Br are exposed to ionizing radiation or maintained at 100°C for an hour. The radiation chemical yields (G values) are respectively 230 and 170 at RT. No “red” phosphorus is formed. The diphosphines react with Br 2 to form CCl 3 Br; with Cl 2 to form CCl 3 Br, Br 2 and PCl 3 ; and with EtOH to form CCl 3 P (OEt) 2 and CCl 3 P (H) (O) (OEt).

Durch Umsetzung von CF 3 CSSH mit BrMn (CO) 5 und BrRe (CO) 5 bei 50° in Dimethyläther gemäß BrM (CO) 5+CF 3 CSSH → CF 3 CSSM (CO) 4 + CO + HBr (M = Mn, Re) gelingt die Darstellung der kovalenten, z. T. sublimierbaren, thermisch und gegen Luftsauerstoff recht stabilen Tetracarbonyl-trifluordithioacetate des Mangans und Rheniums. Abstract

Hydrogen chloride, cyclohexylchloride and the new compound C 4 Cl 5 -C 6 H 11 (cyclohexylpentachlorobutadiene) are formed when mixtures of hexachlorobutadiene and cyclohexane are exposed to γ-radiation or 2 MeV-electrons from an accelerator. The mass- and IR-spectrometric analyses of the new product indicate that the C 6 H 11 -group is bound mainly to a primary C-atom of the hexachlorobutadiene. The 100 eV-yield of C 4 Cl 5 - C 6 H 11 is of the order of 3.3 ± 0.15 molecules/100 eV. Because hexachlorobutadiene is a by-product in the technical synthesis of chlorine containing hydrocarbons' new reactions of this compound could be of industrial importance. A mechanism is formulated to explain the formation of the products.

Durch Umsetzung eines Gemisches von Frangulin A und B bzw. reinem 1.6.8-Trihydroxy-3methyl-anthrachinon-6-O-α-ʟ-rhamnosid (Frangulin A) mit α-Acetobromglucopyranose nach einer modifizierten Koenigs-Knorr- Methode wurde Glucofrangulin A erhalten und über das Acetat identifiziert. Das synthetische Glucofrangulin-A-Acetat ist in jeder Hinsicht mit dem von SEEBECK und SCHINDLER sowie MÜHLEMANN aus natürlichem Glucofrangulin hergestellten Acetat identisch. Durch NMR-Vergleich mit verschiedenen Anthrachinon-1- bzw. -8-glykosiden wird für das Glucofrangulin A die endgültige Struktur eines 1.6.8-Trihydroxy-3-methyl-anthrachinon-6-O-α-ʟ-rhamnopyranosyl-8-O-β-ᴅ-glucopyranosides bewiesen.

Abstract

An thymektomierten, scheinoperierten und normalen NMRI-Mäusen wurde mit 3 H-Thymidin autoradiographisch die Mesothelzell-Proliferation am Omentum majus nach intraperitonealer Stimulierung mit Schaferythrozyten untersucht. Bei thymektomierten Tieren war die Mesothelproliferation am 1. und 2. Tag nach der Injektion im Vergleich zu scheinoperierten Tieren hochsignifikant vermindert. Bei normalen Tieren war die Mesothelproliferation nach Sekundärstimulus geringer als nach Primärstimulus. Diese Befunde und ihre Deutung werden diskutiert.

Chicken insulin was tested in mice in vivo and in vitro. The biological activity was compared to bovine insulin (=100%) as follows: 1. In vitro: Fat pad test 375%; 14 C incorporation into the lipids of fat pads 369%: hemidiaphragm insulin assay 373 percent. 2. Localized intraperitoneal action in vivo: 14 C incorporation into the fat pads 195%: U- 14 C-glucose incorporation into glycogen of hemidiaphragm 215 per cent. 3. Mouse convulsion test: The activity was slightly increased.

Abstract

In order to test recent interpretations of field indicating absorption changes in photosynthesis, the influence of the electric field on the absorption spectrum of chlorophyll-b is measured in vitro. 50 monomolecular layers of chlorophyll-b are built up on quartz slides by the technique of Blodgett and Langmuir. Absorption changes induced by an electric field linearly increasing from −1.8 · 10 6 V/cm to +1.8 · 10 6 V/cm are detected by a repetitive photometer. The absorption changes can be separated into one part, which is proportional to the electric field strength, and a second one, which is proportional to the square of it. The linear change of absorption in the region at ~650 nm may be interpreted as a shift of the absorption band of about .

Abstract

A selective concentration of “Panton-Valentine”-leukozidine (PVL) from the culture-supernatant Staphylococcus aureus was achieved through adsorption of most of the a-hemolysin, coagulase, eggyolk-opacity factor and fibrinolysin to aluminiumoxide cholesterol. PVL was further concentrated through dialysis against a saturated ammonium sulfate solution and subsequent pressure-filtration. Through pressure-filtration relatively low-molecular staphylococcal substances, such as a part of a nuclease, could be separated from PVL.

Abstract

Fluorescense spectra of lactate dehydrogenase * (E.C. 1.1.1.27) were investigated in the presence of the coenzyme fragments dihydronicotinamide mononucleotide and dihydronicotinamide-ribose-5'-pyrophospho- (P 2 ) -5“-ribose. The reduced mononucleotide is enzymatically less active as a hydrogen donor. However, formation of a complex with the enzyme was not observed under the conditions used. All the other substances: dihydronicotinamide-ribose-5'-pyrophospho- (P 2 ) -5“-ribose, dihydronicotinamide- benzimidazole-dinucleotide, dihydronicotinamide-3-desazapurine-dinucleotide and dihydronicotinamide-6-mercaptopurine-dinucleotide form more or less stable complexes with lactate dehydrogenase. The complexes do not markedly differ from the complex formed with the natural cofactor. In all cases spectra indicate change in conformation of the coenzyme by forming the coenzyme-enzyme-complex which has been proposed by VELICK 1 too. The cysteine residues of the lactate dehydrogenase are not essential for binding the coenzyme to the active center; this was shown with mercury blocked enzyme.

Abstract

The colloidal dye lithium carmine was added in vitro to normal human serum. Electrophoretic experiments showed that the dye was associated mainly with α 2 -globulins, small amounts with the albumin and only traces with the γ-globulins. The main complex was eluted with the macroglobulin peak obtained by gel filtration on Sephadex G-200 and sedimented in the heavy fraction on density gradient ultracentrifugation. The dye-protein complex could be precipitated with an antiserum specific for a 2 -macroglobulin. Gel filtration of a solution of pure a 2 -macroglobulin, to which lithium carmine was added, demonstrated that the dye was bound to this protein.

Abstract

Der Häutungszyklus der Strandkrabbe wird mit Hilfe neuer Kriterien in 21 verschiedene Stadien eingeteilt. Als Charakteristika für die einzelnen Stadien dient neben den bisher angewendeten morphologischen Merkmalen der Entwicklungszustand der Regenerate und die Geschwindigkeit, mit der die Krebse den Häutungszyklus durchschreiten. Für jedes Stadium wird der Häutungshormongehalt mehrerer Tiere bestimmt und eine Hormontiterkurve für einen Häutungszyklus aufgestellt. Aus den Änderungen des Hormongehaltes und den gleichzeitig stattfindenden morphologischen und physiologischen Veränderungen wird auf die Funktionen des Häutungshormones geschlossen. Vor der Häutung erfolgen zwei Hormonausschüttungen, die eine ca. 6 - 8 Tage, die andere 1 - 2 Tage vor der Häutung. Die erste Hormonausschüttung löst wahrscheinlich die Apolyse aus, die zweite bewirkt eine Erhöhung des osmotischen Wertes der Hämolymphe und führt damit zur Auslösung der Häutung im engeren Sinne. Unmittelbar nach der Häutung wird erneut Hormon ausgeschüttet, das vermutlich die Härtungsprozesse in der Cuticula steuert. Nachdem sämtliche mit der vorangegangenen Häutung zusammenhängenden Prozesse abgeschlossen sind, löst eine weitere kurzfristige Hormonausschüttung die Regeneratentwicklung und vielleicht den nächsten Häutungszyklus aus. Ein Vergleich des Häutungshormongehaltes der Strandkrabbe während eines Häutungszyklus mit demjenigen von Insekten während der Puppenhäutung ergibt einige Übereinstimmungen, z. B. in der Höhe des Hormongehaltes und einige Unterschiede, die sich aus der unterschiedlichen Lebensweise dieser Tiere erklären.

Abstract

The tubular peritrophic membranes of Calliphora are formed by the secreting cells of the proventriculus at a rate of 3.5 ± 1 mm per hour. It has been estimated that each of these cells produces its own weight in membrane material in about one day. Autoradiography of peritrophic membranes from flies injected with glucose-C 14 show cross bands as well as longitudinal bands, indicating a periodic incorporation of radioactive material and therefore a different chemical composition, which may be of importance for transport phenomena.

Abstract

The synthesis of plastidquinones was studied during the development through the degeneration of Ficus leaves and compared with chlorophyll and carotenoid synthesis. 1. The plastidquinone content of Ficus chloroplasts increases steadily with increasing age of leaf tissue. The augmentation of the plastidquinone level is mainly due to the synthesis of the reduced quinone forms plastoquinol 45 (Plastohydrochinon 45) and a-tocopherol, the chromanol of α-tocoquinone. In several years old leaves the concentration (moles) of plastoquinol 45 and α-tocopherol reaches or exceeds that of chlorophyll a. 2. The naphthoquinone vitamin K 1 (phylloquinone) as well as the oxidized quinone forms (α-tocoquinone and plastoquinone 45) seem not to be produced in excess amounts. The levels of the latter increase, however, in older and senescent leaves, apparently by oxidation of some α-tocopherol and plastoquinol 45 respectively. 3. The production of excess plastidquinones starts very early during leaf development. The main synthesis phase begins after the end of chlorophyll and thylakoid synthesis and continues in senescent Ficus leaves during the breakdown of chlorophyll until the early yellow stage. 4. The young pale green leaves still rolled up within the buds, possess a quinone and carotenoid composition which is quite different from that of fully expanded leaves. It resembles that of shortly illuminated etiolated seedlings. 5. The carotenoid level of older fully grown Ficus leaves slowly increases, mainly by synthesis of lutein. These additional carotenoids represent excess amounts, which are deposited together with the excess plastidquinones in the plastoglobuli of the plastid stroma. 6. There is no indication for a re-incorporation of excess plastidquinones in the general chloroplast metabolism. Thus the excess plastidquinones, accumulated in the osmiophilic plastoglobuli, may be regarded as secondary plant products.

Abstract

Chloroplasten-Ribosomen (67s) von Allium porrum besitzen eine niedermolekulare, einheitliche RNS-Komponente mit einer Sedimentationskonstanten von 5s. Die 5s-RNS der Chloroplasten-Ribosomen wird nach Chromatographie an methyliertem Serumalbumin mit einer höheren NaCl-Konzentration von der Säule eluiert als die entsprechende 5s-RNS der Cytoplasma-Ribosomen. Auf Grund der gleichen Sedimentation und des einheitlichen Verhaltens bei Gelfiltration an Sephadex G-100 kann auf eine ähnliche wenn nicht gleiche Molekülgröße der 5s-RNS aus Cytoplasma- und Chloroplasten-Ribosomen geschlossen werden. In grünen und etiolierten Pflanzen von Zea Mays werden neben Cytoplasma-Ribosomen auch Plastiden-Ribosomen nachgewiesen. Beide Ribosomenklassen besitzen neben ihren hochmolekularen RNS-Komponenten jeweils eine 5s-RNS. Damit kann ganz allgemein gesagt werden, daß Plastiden-Ribosomen, unabhängig vom Entwicklungszustand der Plastiden, 3 RNS-Fraktionen besitzen: 5s, 16s, 23s.
